Gettting the following error on the web page for LS 4.0.0.x (March 12, 20101 buil)
The current identity (NT AUTHORITY\SYSTEM) does not have write access to 'C:\Windows\Microsoft.NET\Framework64\v2.0.50727\Temporary ASP.NET Files'

Though this did work, it should be noted that the folder this needs to run under for x64 is the following.
C:\Windows\Microsoft.NET\Framework64\v2.0.50727

Your default permissions for that folder do not seem correct/default. A fresh IIS install lists:
Everyone: FC
CREATOR OWNER: special
SYSTEM: FC (inherited)
Administrators: FC (inherited)
Users: RX
IIS_IUSRS: RX (inherited), M
TrustedInstaller: special
Everyone: FC
CREATOR OWNER: special
SYSTEM: FC (inherited)
Administrators: FC (inherited)
